Going old school Lita Ford and Doro were the best.
In the new-er bands I like Maria Brink a lot. She doesn't fry scream, which is cool with me. That is overdone most of the time. A little is okay for some extra flavor, but it cant be the whole meal.
Nita Strauss is an excellent guitarist. Powerful and melodic.
Tina S is another incredible guitarist. She has truly amazing technical ability but isn't one for the spotlight. She plays for the sheer joy of playing.
There are a ton of Japanese female metal bands out there and they have some pretty amazing talent.
An honorable mention: Samantha Fish is a fabulous guitarist and singer but not metal, old school blues. Very cool.
