It is the saying in the text, "Therefore whosoever slayeth Cain, vengeance shall be taken on him sevenfold." that is itself the non-corporeal sign, the 'warning, which serves to prevent Cain from being killed. YHWH'S words to Cain and the sign are not two different things, however closely related but one and the same thing; YHWH'S words are first spoken to Cain and are then redescribed as a protective sign.

I don't think the descendants of Cain were all killed off.
He placed a mark (sign) on Cain so those who found him wouldn't kill him.
