There aren't many movies that choke me up, but as a kid, I remember a particular scene in The Call of the Wild (1972) with Charlton Heston.
It's the bet... I really struggle to hold back the emotions with this one.
I couldn't find just the scene, so here's the full movie. Go to 01:28:00. The build up starts before, but watch through to the end of the bet.
The movie ending is powerful, too, but I still prefer this part.
